The posterior tibial tendon (PTT) is very vulnerable and insufficiency was recognized as the major cause of adult acquired flat foot. Some patients have no predisposing cause clinically recognized, showing that the characteristics of the individual plays an important role in tendinopathy. This study investigated whether genetic variants in matrix metalloproteinase (MMP) are associated with dysfunction PTT.
METHOD
One hundred women who served dysfunction PTT, the histopathological examination of the tendons and magnetic resonance imaging (MRI) confirms tendinopathy, and 100 women without symptoms who PPT intact as assessed by MRI and a control group, were evaluated for MMP-13 G-77 A> > G (rs2252070) polymorphism, individually and in haplotypes, as well as in combination with MMP-1 G-519 A >> G (rs1144393), MMP-1 G-1607 G >> GG (rs1799750) and MMP-8 G. 799 C >> T (rs11225395) polymorphism with PTT dysfunction.
RESULTS
The frequency of allele A (MMP-13 G-77 A >> G (rs2252070) polymorphism) was significantly higher in case group (76% and 61%, respectively; p = 0.010, odds ratio = 2.02; 95 % confidence interval = 1.32 to 3.12). The distribution of genotypes also differed significantly between groups (p = 0.001, odds ratio = 2.82; 95% confidence interval = 1.58 to 5.02). global haplotype analysis showed a significant difference between the two groups.
CONCLUSION
In conclusion, these findings suggest that MMP-13 G-77 A >> G (rs2252070) polymorphism individually, as well as haplotypes MMP-1 G-519 A >> G (rs1144393), MMP-1 g.- 1607 G> > GG (rs1799750) and MMP-8 G-799 C >> T (rs11225395), can cause dysfunction PTT.
NanoSim: sequence based nanopore reading simulator statistical characterization.
The Minion sequencing instrument of Oxford Nanopore Technologies (ONT) produces long read length of single-molecule sequencing – a valuable feature for a detailed characterization of the genome. To realize the potential of this platform, a number of groups that develop bioinformatics tools tuned to the unique characteristics of the data. We note that these development efforts will benefit from the software simulator, the output can be used to benchmark analysis tools.
Here, we introduce NanoSim, fast and scalable read simulator that captures a specific technology ONT data features and allows for adjustments to the improvement of nanopore sequencing technology. The first step is to read the NanoSim characterization, which provides a comprehensive analysis of the alignment-based and generates a set of read profile serves as the input for the next step, the simulation stage. Phase simulations using a model built in the previous step to generate in silico reading for a given reference genome. NanoSim written in Python and R.

In this work, we model the basic error-call from ONT to read to inform simulation sequence with similar characteristics. We show the performance NanoSim on publicly available datasets generated using chemical R7.3 and R7 and different sequencing kit and compare the resulting synthetic sounds with other old-order simulator and experimental ONT read. We hope NanoSim have a role that allows the field and the benefits of the development of next-generation technology for nanopore sequencing scalable long reads, including genome assembly, mutation detection, and even a metagenomic analysis software.
